In the office

'In the office' is used to talk about the physical presence of someone or something in a workplace which is referred to as the office.

 Have a look:

He spends most of his time in the office.

(Here, we mean that the subject is most of the time physically present at the office location.)

At the office

We use 'at the office' to talk about the concept and the idea of someone's workplace. We often use 'at work' instead of 'at the office' in this context. For example:

 

He spends most of his time at the office.

( Here, we mean that the subject spends most of their time working.)
